Opening the X-files: Exploring HBx protein’s role in transcriptional enhancement |
Description |
Background: HBV’s cccDNA is a compact stable viral form. For transcription to occur, the cccDNA needs to open up to expose its promoters for recruitment of host proteins. Promoter-targeting therapies may be enhanced if the cccDNA can be “activated”. HBV produces a multifunctional X protein (HBx), that plays a key role in transcriptional activation. Previous studies show HBx supplementation enhances HBV replication, but the degree to which this can be pushed has not been analyzed. Purpose: To determine whether and to what degree excess HBx in a cell culture model can enhance the HBV transcription. Methods: HepG2-NTCP subclone cells were co-transfected with a plasmid encoding HBx and either a wildtype HBV plasmid or mutant (HBx-null). Gene expression was measured with luciferase assays, and viral markers were measured by qPCR and Western blot. Conclusion: This work offers deeper insight into HBx's mechanisms and a route for potential therapies targeting the promoter region. |
