G4Q-binding ligands: a starting point for potential cure therapies for chronic hepatitis B infections |

Introduction: HBV’s chronicity is partly due to the virus’ cccDNA. Current drugs target processes downstream of cccDNA, but not the cccDNA itself. A highly conserved segment in the preC promoter region of cccDNA, known as a G-quadruplex (G4Q) was reported. G4Q-binding ligands are well explored in cancer. Purpose: To study the effects of two G4Q-binding ligands, TMPyP4 and Braco19, on HBV replication. Methods: AlamarBlue assay was performed to test the optimal drug concentration in the HepG2-NTCP cell model. Infection studies were performed using HepAD38-derived HBV and viral and cellular markers were analyzed by HBsAg ELISA, HBcAg immunofluorescence, and qPCR. Results: Infection was confirmed in the cell culture model and the effects of Braco19 and TMPyP4 on HBV replication were quantified. Off-target effects were quantified using a panel of housekeeping genes. Conclusion: G4Q-binding ligands may act as a reasonable starting template for potential future HBV therapeutics. |
