Background: Chronic HBV persists in cells in a form of cccDNA that incorporates a knot-like structure formed by G-rich sequences, a G-quadruplex(G4Q). G4Q in the pre-core promoter region is vital for viral transcription. Mithramycin and daunorubicin are FDA approved anti-cancer drugs that can bind to G4Q and displace transcription factors preventing transcription. Purpose: This project aims to measure the efficacy of those drugs against HBV. Methods: The optimal drug dose in HepG2-NTCP cells was determined with Alamar blue viability assay. Next, using an HBV pre-core promoter luciferase plasmid, the impact of the drugs on luciferase output was measured. Finally, drugs were tested in an HBV infection model. Results: Both drugs have inhibitory effects on the pre-core promoter of HBV in luciferase model. Infection studies will follow. Conclusion: These drugs are suggested to block the binding of transcription factors, and therefore can act as a blueprint for a novel HBV drug. |
